Abstract
Background. Lactoferrin, in addition to having antimicrobial effect and protect the surface tissues of the mouth, teeth and mucous, it was also believed to induce osteoblastic activity and inhibit osteoclastic activity. However, there was no study has tested the role of lactoferrin in increasing bone formation activity in the healing stages. Therefore the objective of our study is to observe the differences of bone formation increasing activity in the topical application of lactoferrin on Wistar rats (Rattus norvegicus) bone defect in the healing stages. Method. Randomized post test only control group design performed in 18 male Wistar rats aged 12 weeks. The defect was made with a 2mm depth drilling in mandible and was applicated 1,5ìm/mL lactoferrin gel. The observation of bone formation activity performed on 3 stages of healing, i.e: day 7, 21, and 42 after drilling. HE staining was conducted to observe
the three-dimensional stereologi trabecular variables, whereas TRAP performed to observe osteoclasts variables. The results of both staining observation was used for histomorfometri analysis. Results. Multivariated ANOVA showed significant differences in Trabecular Volume Percentage (Tb.BV/TV) (p<0.0001), Mean Trabecular Plate Density (MTPD) (p<0.0001), Mean Trabecular Plate Separation (MTPS) (p <0.0001), and Active Surface resorption (ARS) (p=0.0012) between the treatment and control groups at each stage of healing. While Mean Trabecular Plate Thickness (MTPT) only significant difference between treatment and control groups
(p <0.0001), but not significantly different at each stage of healing. Tb.BV/TV variable values, MTPT, and MTPD in the treatment group at each stage of healing is smaller than the control group. In contrast, the value of MTPS and ARS in the treatment group at each stage of healing was greater in the control group and increased in each of the stages. Conclusion. Topical application of lactoferrin enhances bone formation activity in all the variables in each stage of healing, especially at this stage of remodeling.
