Abstract


A  research  on  in  vitro  fresh  sperm  preparation  for  maintaining  sperm  viability  at storage temperature of 14°C using tannin supplementation of lamtoro leaves (Leucaena leucocephala)had  been  carried  out  in  both  Physiology  and  Animal  Reproduction  Laboratory,  Faculty  of  Animal Science, Universitas Gadjah Mada for  fresh  sperm  preparation and sperm  quality evaluation, during March to July of 2009.The experiment was designed using the Complete Random Design consisted of 5  treatments  and  5  replications.  Variables  measured were  viability,  motility  and  morphology  of  the treated fresh sperm stored for 14 days at temperature of 14°C. Based on the analysis of variance, the treatments  had  significant  effect  (P<0.05)  on  viability,  motility  and  morphology  of  the  sperm.  In terms  of  viability,  P1(64.07%±0.45)  it  was  higher than  control  (P0=26.71%±0.54),  but  it  was  not different  for  P2(57.44%±0.52),  P3(49.49%±0.71), and  P4  (47.83%±0.61).  The  progress  and  very progress  motility  of  spermatozoa  were  only  found  in control,  and  not  in  others.   Non-progressive spermatozoa proportion of P1, P2, P3 and P4,was found respectively. They were significantly higher than control.For the  morphology, P4 had lower normal spermatozoa percentage than P0, P1, P2 and P3,  whereas among P0, P1, P2, and P3, there were not different (P>0,05).
