Abstract


Chronic stress may cause morphological changes in the hippocampus following by deficit in hippocampal
function including the impairment ofworkingmemory.
The aimof this study is to investigate the changes the thickness of pyramidal layer CA1 hippocampus and it’s
correlation with working memory of the rats after chronic stress.
The subjects of this study were 10male rats (Rattus norvegicus) divided into control group (C) and treatment
group (T), each group consist of 5 rats.Workingmemory performancewas tested for each group by using 8-arm
radial maze for 12 days. Working memory measured by error type A. After tested, T rats group were given
chronic stress procedure with electrical footshock of 60 volt for 10 seconds with 15 seconds interval for 15
minutes each day for 25 days.After chronic stress, the workingmemory of T and C group were tested again by
using 8-armradialmaze for 12 days.At the end, the ratswere killed and the brainwas removed tomake paraffin
blocks of CA1 hippocampus to stain with hematoxylin. The thickness of pyramidal layer CA1 hippocampus
measured undermicroscope at 400xmagnification.
The results of this study showed that chronic stress caused decreasing of workingmemory of the rats showed
by error typeAforT group are higher thanCgroup (p<0.05).Chronic stress also caused morphological changes
in hippocampus showed by the thickness of pyramidal layer ofCA1 hippocampus in T group are smaller than C
group (p<0.05). With correlation product moment test there are positif correlation between the thickness of
pyramidal layer of CA1 hippocampus with workingmemory of the rats after chronic stress (p<0.05).
The study concludes that chronic stress caused decreasing of workingmemory of the rats and the thickness of
pyramidal layer of CA1 hippocampus. It is concerned there are positive correlation between the thickness of
pyramidal layer of CA1 hippocampus with workingmemory of the rats after chronic stress.
